- COMPOSITION:
Chloroquin diphosphate 50 mg/l ml (= 31 mg Chloroquin base);
64.5 mg/l ml (= 40 mg Chloroquine base) in ampoules of 2 ml and
5 ml and 250 mg in one tablet.
ACTION:
A synthetic drug, quickly absorbed in the digestive tract and is
partially bound to serum proteins. Its action is associated with the
effect on the cellular membranes, as well as with the effect on cell
proliferation. It destroys the erythrocytic forms of the malarial
plasmodium, some protoza, amebae, the causative agents of
fascioliasis. It is still not certain which of its numerous actions
is responsible for the effect of arthrochin in rheumatic diseases.
It is fixed in the tissues, mainly in the reticuleondothelial
system: liver, spleen muscles, eye, etc.; it is selectively
accumulated in considerably higher concentrations in the inflamed
tissues. It is metabolized in the liver and is very slowly excreted.
INDICATIONS:
Malaria (therapeutically and prophylactically), amebic hepatitis,
abscess in the liver, lambliasis, chronic articular rheumatism,
chronic rhematoid arthritis, spondyliis, lupus erythematosus, acne
rosacea, lichen ruber planus, trematode invasion, etc.
CONTRAINDICATIONS:
Diffuse hepatic and renal diseases, severe cardiac diseases with
rhytmic disorders, diseases of the central nervous system,
leukopenia, susceptibility to spasms of the stomach and intestines.
SIDE EFFECTS:
Eye lesions are most frequent: retinopathy manifested with blurred
vision, disturbances in color vision; reversible depositions in the
cornea. Gastric disturbances (nausea, vomiting, anorexia) have also
been observed, as well as rarely - nervous manifestations
(dizziness, tinnitus, insomnia, headache). Hepatic injuries are
possible, as well as myocardial dystrophy, dermatitis, leukopenia. |
PRECAUTIONS:
Blood count to be followed up each month. Regular ophthalmological
contol is necessary for the early detection of eye alterations -
every 6 months.
DRUG INTERACTIONS:
Its plasma concentration can be increased by simultaneous
administration with penicillamine, and its toxic effects can be
enhanced.
DOSAGE AND ADMINISTRATION:
Adults - initial dose of 2 tablets daily, taken every 12 h is
recommended for the first 2-4 weeks, subsequently transferred to a
maintenance dose of 1 tablet daily for months for the treatment of
rheumatoid arthritis.
Children - the treatment starts with 1/8 to ½ tablets daily,
depending on age.
For malaria treatment, prophylactically - 2 tablets once weekly, and
therapeutically for adults - 4 tablets on the first day, at an
interval of 6-8 h. On the following days a single tablet is
administered.
Special schedules are applied for the other diseases indicated for
arthrochin treatment.
Parenterally - up to 10 ml from the solution must be injected deeply
intramuscularly and the patient should remain lying for 15-30 min
after the injection.
MEDICINAL FORM AND PACKAGE:
Packages with 10 and 50 ampoules of 2 ml and 5 ml (5 % and 6.45%).
Packages with 100 filmtablets.
STORAGE:
At moderate temperature (15-30° C) in a place protected from light.
EXPIRY:
Ampoules - 3 (three) years.
Filmtablets - 2 (two0 years.
