- COMPOSITION:
Chlorpromazine hydrochloride 50 mg/ 2 ml, 25 mg/5 ml, 50 mg/5 ml
and 100 mg/5 ml in one ampoule.
ACTION:
A neuroleptic with phenothiazine structure. It has adrenolytic and
cholinolytic action. Inhibits the reuptake of the transmitters
epinephrine, dopamine and serotonine in the presynaptic segment of
the neuron and enhances their turnover. It has a sedative effect on
the higher segments of the central nervous system, which does not
induce sleep or narcosis. Eliminates psychic strain and fear. It has
an antipsychic action. In high doses it causes extrapyramidal
disorders. Potentiates the effect of pain-relieving, local
anesthetic and hypnotic agents. It decreases body temperature. The
drug has an antiemetic action and inhibits the hiccup refles.
PHARMACOKINETICS:
Good gastrointestinal absorption. Binds with plasma proteins up to
91-99 %. Its plasma half-life is 16-20 h. the drug is metabolized in
the liver and is excreted with the urine - metabolized and unchanged
in about 6 %.
INDICATIONS:
It is widely used in many fields of the clinical practice:
Internal medicine: vomiting of various origin; persistent pain in
combination with analgesics; severe intoxications with carbon
monoxide; severe attacks of asthma and singultus.
Pediatrics: toxicosis in breast-fed infants, acute encephalitis,
acute pneumonia with dyspersia, vomiting, persistent cough
paroxysms.
Obstetrics and gynecology: eclampsia, vomiting during pregnancy.
Psychiatry and neurology: schizophrenia and cyclophrenia, involutive
psychosis, generalized paroxyms of epilepsy, chorea minor.
Surgery: premedication for narcosis, prophylaxis and treatment of
traumatic and postoperative shock.
Dermatology: refractory pruritus, etc. |
CONTRAINDICATIONS:
Hepatic diseases, advanced renal insufficiency, disordered function
of hematopoietic organs, decompensated cardiac diseases,
thrombembolic disease; comatose states and acute cerebral trauma;
ulcer disease of the stomach and duodenum; intoxiactions with
barbiturates, narcotics and alcohol.
SIDE EFFECTS:
Reduction of systolic and diastolic pressure is likely to occur at
the beginning of the treatment that can lead to dizziness and
lassitude. Tachycardia, dryness in the mouth, sense of heaviness in
the head can sometimes be observed.
PRECAUTIONS:
Do not prescribe to drivers! Alcohol is forbidden during the
treatment with the drug, it should be prescribed cautiously with
hypnotics, barbiturates (incompatible for simultaneous
administartion), other psychotronic agents.
DRUG INTERACTIONS:
Chlorazin reduces the action of coumarin anticoagulants, insulin,
norphenephrin, metformin and sulfanylurea antidiabetic agents.
Chlorazin reduces the effect of lithium salts. Propranolol elevates
the level of chlorpromazine in plasma.
DOSAGE AND ADMINISTRATION:
Parenterally applied: intramuscularly and intravenously.
Intramuscularly injected in a dose of 0.025-0.05 g.
Intravenously injected in a dose of 0.025-0.05 g and due to the acid
reaction of the solution, it should be diluted in infusion fluid in
advance.
MEDICINAL FORM AND PACKAGE:
Packages with 10 and 50 ampoules of 50 mg/2 ml, 25 mg/5 ml, 50 mg/5
ml and 100mg/5 ml.
STORAGE:
At moderate temperature (15-30 C), in a place protected from light.
EXPIRY:
