- COMPOSITION:
Pentoxyfylline 100 mg in an ampoule of 5 ml
ACTION:
Pentofyllin is an alkyd derivative of theophylline and it possesses
a marked vasodilatory action, without the central effect of
theophylline. It belongs to the group of vasodilators with myotropic
effect, acting mainly on the cerebral vessels. Pentophylline
inhibits the phosphodiesterase and increases the cAMP concentrations
in the smooth muscle cells, resulting in increased binding affinity
to the cell membrane, which in turn leads to the stabilization of
the latter and the lower free calcium levels in the cell.
Pentophyllin increases the cerebral blood flow. It acts as a
thrombocyte antiaggregation agent by rising the cAMP level in the
thrombocyte, the prostacycline level and by reducing the viscosity
and flexibility of the erythrocyte membrane.
PHARMACOKINETICS:
Pentophyllin is well absorbed and uniformly spread in the body.
Maximum plasma concentrations are reached 30 min after oral
administration and are maintained for 4-6 h.
INDICATIONS:
Pentophyllin is indicated for treatment of all cases of
cerebrovascular insufficiency, both acute and chronic, in young and
elderly patients. It is used for treatment of angiopathy of
diabetic, atherosclerosic, or inflammatory origin, as well as for
treatment of angioneuropathy, paresthesia, acrocyanosis, Raynaud's
disease, postthrombosis syndrome, cerebral atherosclerosis
(dizziness, headache), etc. |
CONTRAINDICATIONS:
Pentophyllin should not be applied intravenously after myocardial
infarction; grave coronary or cerebral sclerosis, pregnancy,
hypotension, or to patients with unstable blood circulation.
SIDE EFFECTS:
In some cases: headache, tremor, sleep disorders, retrosternal pain,
lower blood pressure, bradycardia or tachycardia, may be observed.
DRUG INTERACTIONS:
The effects of insulin, metformin and sulphanylurea antidiabetic
agents are potentiated after high parenteral doses of Pentophyllin.
DOSAGE AND ADMINISTRATION:
For bed-ridden patients, a single dose of 100 mg (1 ampoule) is
administered slowly (5 min) intravenously or intraarterially 100-300
mg by infusion.
MEDICINAL FORM AND PACKAGE:
Ampoules of 5 ml, containing 100 mg (2 %) in packs of 50 and 100.
STORAGE:
At room temperature (15-30° C) in a place protected from direct
sunlight.
EXPIRY:
3 (three) years.
